Murine Terminal Deoxynucleotidyl Transferase
نویسنده
چکیده
Terminal deoxynucleotidyl transferase (TdT) 1 is an enzyme which has the unique property of polymerizing deoxyribonucleotides onto a primer in the absence of a template (1-4) . Chang (4) first reported that TdT is present only in the thymus among various avian and mammalian tissues examined . Her finding was extended to humans by McCaffrey et al . (5), who found that the enzyme was restricted to the thymus in normal tissues but was found in peripheral blood lymphocytes from a patient with acute lymphoblastic leukemia. The specific association of TdT with the thymus has led to the suggestion that TdT might act as a somatic mutator andcould be implicated in the generation of immunological diversity (6) . Since TdT was thought to be restricted to the thymus, its distribution among thymocytes has been a critical problem . At least two major thymocyte subpopulations have been identified in mice and other animals (7-13) . The major subpopulation is immunoincompetent, cortisone-sensitive, rich in Thy, TL, Gix, and Ly antigens, but bears little or no H-2 antigen . The minor subpopulation, on the other hand, is immunocompetent, cortisone-resistant, rich in H-2 but low in Thy and Ly antigens, and carries no detectable TL or G, x. Whether the minor subpopulation is derived from the major one is still debatable (12) . In this communication we present evidence for two forms of TdT in murine thymocytes and for their distribution between two thymocyte subpopulations . Also, the occurrence of TdT in murine bone marrow cells is reported .

Track detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)
Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...
متن کاملTerminal deoxynucleotidyl transferase is found in prothymocytes
Terminal deoxynucleotidyl transferase is an enzyme which has the unique property of polymerizing polydeoxynucleotides onto a primer in the absence of a template (1,2). This enzyme is found both in the thymus and the bone marrow of birds, rodents, and humans (3-7). Whether the marrow cells that contain terminal transferase are related to thymocytes, or are on a separate pathway of differentiatio...
متن کاملTerminal deoxynucleotidyl transferase: the story of a misguided DNA polymerase.
Nearly every DNA polymerase characterized to date exclusively catalyzes the incorporation of mononucleotides into a growing primer using a DNA or RNA template as a guide to direct each incorporation event. There is, however, one unique DNA polymerase designated terminal deoxynucleotidyl transferase that performs DNA synthesis using only single-stranded DNA as the nucleic acid substrate. In this...

Heparin inhibited the activities of: (a) reverse transcriptase from simian sarcoma virus; (b) DMA polymerases a, (i, and y from phytohemagglutinin-stimulated normal hu man lymphocytes, human leukemic lymphoblasts, and Molt-4 cells which are of acute lymphoblastic leukemia origin; and (c) terminal deoxynucleotidyl transferase from the two latter cell types. A 10-fold excess of enzyme overcame he...
متن کاملOccurrence of a terminal deoxynucleotidyl transferase-like activity in N-2-fluorenylacetamide-treated rat liver.
A new enzyme activity that resembles terminal deoxynucleotidyl transferase (TdT-like activity) was found in rat liver. Upon treatment of rats with a hepatocarcinogen, N-2-fluorenylacetamide, the TdT-like activity increased in parallel with DNA polymerase Ci and 'Y. By phosphocellulose column chromatography, the TdT-like activity was separated from DNA polymerase Ci, fJ, and 'Y. With a poly(dA)-...
